    In chronic inflammatory conditions such as renal cancer, MDSCs appear and suppress the cancer immune response. We have previously established a method for analyzing MDSCs in the peripheral blood of patients with renal cancer using the expression level and coefficient variation of GPI-80, combining with myeloid-differentiation markers. We investigated changes in the number and function of MDSCs using peripheral blood before and during cancer therapies.
    The increasing MDSCs indexes (CD16 and LAP-1 expression level) were associated with poor response to each cancer therapy. In cases of immune-related side effects, GPI-80 coefficient variation, which is other MDSCs indicators, was reduced. In the case of complete response, neutrophils with increased GPI-80 expression level were significantly observed before the therapie. Since GPI-80 was involved in the activation of NF-κB, it is possible that it is involved in neutrophil function that enhances the cancer immune response.

    The chronic inflammation caused by obesity is thought to be the underlying pathology of type 2 diabetes and its complications. Elucidation of the mechanism of inflammation-induced insulin resistance is important in the prevention of intervention in metabolic syndrome. We establish a novel animal model for non-obese diabetes and uncover a link between glucose metabolism and inflammatory response.
    5-Aminolevulinic acid (5-ALA) is a natural amino acid and the first precursor of heme biosynthesis. 5-ALA or heme is associated with glucose metabolism in vivo. We previously showed that 5-ALA synthase ALAS1 heterozygous (Alas1+/-) mice displayed insulin resistance, which was cured by 5-ALA administration.
    Here, we test whether 5-ALA have any potential effect on innate immune system. Alas1+/- mice exhibited reduced inflammatory responses and 5-ALA administration increased ROS production in neutrophils. These findings show that 5-ALA is a potential therapeutic target against metabolic syndrome.

    Superoxide dismutase 1 (Sod1) plays pivotal roles in antioxidation via accelerating the conversion of superoxide anion radicals into hydrogen peroxide, thus inhibiting the subsequent radical chain reactions. While Sod1 deficient cells inevitably undergo death in culture conditions, Sod1-knockout (KO) mice show relatively mild phenotypes and live approximately two years. We hypothesized that the presence of abundant levels of ascorbic acid (AsA), which is naturally produced in mice, contributes to the elimination of reactive oxygen species (ROS) in Sod1-KO mice. To verify this hypothesis, we employed mice with a genetic ablation of aldehyde reductase (Akr1a), an enzyme that is involved in the biosynthesis of AsA, and established double knockout (DKO) mice that lack both Sod1 and Akr1a. Supplementation of AsA (1.5 mg/ml in drinking water) was required for the DKO mice to breed, and, upon terminating the AsA supplementation, they died within approximately two weeks regardless of age or gender. We explored the etiology of the death from pathophysiological standpoints in principal organs of the mice. Marked changes were observed in the lungs in the form of macroscopic damage after the AsA withdrawal. Histological and immunological analyses of the lungs indicated oxidative damage of tissue and activated immune responses. Thus, preferential oxidative injury that occurred in pulmonary tissues appeared to be primary cause of the death in the mice. These collective results suggest that the pivotal function of AsA in coping with ROS in vivo, is largely in pulmonary tissues that are exposed to a hyperoxygenic microenvironment.

  • Enhancement of anti-tumor immunity with membrane-bound interleukin 12 (IL-12),2018.04 - 2021.03,Enhancement of anti-tumor immunity with membrane-bound interleukin 12 (IL-12)

    IL-12 is an important cytokine for the induction of Th1 cell-mediated immunity. There are secretory and membrane-bound types of IL-12, and we have reported that membrane-bound IL-12 exhibits stronger Th1 inducibility than the secretory type. Therefore, we thought that regulation of membrane-bound IL-12 expression might promote anti-tumor immunity mediated by Th1 response. To this end, we first attempted to elucidate the secretory mechanism of membrane-bound IL-12, and found that a proteolytic enzyme called ADAM17 may be indirectly involved. Membrane-bound IL-12 is expected to be associated with some molecule that is cleaved by ADAM17 and is currently being searched for that molecule.

    In renal cancer patients, immunosuppressive neutrophils called myeloid-derived suppressor cells (MDSCs) appear due to cancerous chronic inflammation. On the other hand, intravesical BCG instillation therapy for non-muscle invasive bladder cancer induces strong inflammation and provides a strong anti-tumor immune response. In this study, we investigated the BCG-induced inflammatory response and tried to find the indicators of neutrophil-like cells associated with anti-tumor immune response. As a result, no increase in MDSCs was observed during the BCG treatment. While, the expression of HLA-II and CXCL10 were increased in neutrophil-like cells in urine. Since it is known that HLA-II and CXCL10 induce activation of the adaptive immune system, it was considered that they could be useful indicators of anti-tumor neutrophil-like cells.

    Granulocyte and monocyte adsorptive apheresis (GMA), an effective therapy for inflammatory disorders, exerts an anti-inflammatory influence by utilizing the biological reaction between blood and cellulose acetate (CA) beads, which are the carriers of the GMA column. Although the biological reaction has an optimum temperature, blood contacts the CA beads below body temperature as GMA is performed in an extracorporeal circulation system. We investigated various soluble factors in blood treated with CA beads at 25 degrees C and 37 degrees C. Here, the optimal temperature for IL-1 receptor antagonist (IL-1ra) release induced by CA beads was 37 degrees C, and IL-6 production from monocytic cells was inhibited by the addition of plasma prepared from the CA bead-treated blood at 37 degrees C, rather than at 25 degrees C. These results indicated that physiological heating of the apheresis carrier augmented the anti-inflammatory reaction in vitro. Thus, heating during GMA may be a new approach for augmenting clinical efficacy.
