In my undergraduate days, I was very fortunate in taking a course in Chronobiology by Prof. Yoshihiko Chiba of Yamaguchi University, one of the pioneers of the field in Japan. In the graduate course at Osaka University, I studied molecular biology. In the early days of my postdoc life, in 1999, before the advent of the molecular models of negative feedback loop of circadian rhythm, I identified cryptochrome (CRY) gene in Drosophila corresponding the protein that was called blue-light receptor at that time (Photochem Photobiol, 1999). After some years of the studies of DNA repair problems with Prof. Akira Yasui (IDAC, Tohoku Univ.), I returned to CRY again, and I made transgenic mice ubiquitously overexpressing mutant CRY1 (cysteine414 of CRY1 was replaced with alanine: the molecular implication of the role of cysteine 414 in CRY1 has been elucidated by Prof. Eva Wolf's group as the intermolecular zinc-coordination site in 2014). The C414A mutant CRY1 Tg mice displayed a unique circadian phenotype that their locomotor free-running periods were very long (around 28 h) with rhythm splitting. Moreover, by analyzing unique features of food-entrained rhythm of the Tg mice, I could propose some new points of view to the mechanisms of food entrainment circadian rhythms (Sleep Biol Rhythms, 2016). In addition, I have demonstrated that the male Tg mice show early onset insulin-secretory defect of diabetes mellitus resembling human MODY (Neurosci Lett, 2009; Eur J Clin Invest, 2010; J Diabetes Investig, 2013), which is presumably caused by senescence-associated secretory phenotype-(SASP-) like characters of pancreatic ß-cell. The details of these successive researches of cryptochrome proteins mentioned above are succinctly described together with other group's achievements in my review article (Okano. S., J Diabetes Res. Article ID: 3459246, 2016). Regarding the succeeding research on the Tg mice, I could propose that the SASP-like microenvironment inside islets might play stimulatory roles in the formation of islet fibrosis and also of unique ductal metaplasia, intra-islet ducts with age (Okano. S. et al., Article ID: 7234549, J Diabetes Res. 2019). Currently I am still struggling with the elucidation of the molecular mechanisms of pancreatic ß-cell dysfunction, the alteration of islet configuration and generation of the possible pre-cancerous lesions (the intra-islet ducts) as well as of the unusual circadian rhythms in the Tg mice.
